First Tracks Bolsters Pipeline with Promising Early-Stage Celiac Disease Data

In a significant development for the immunology landscape, biotechnology firm First Tracks announced positive early-stage clinical trial results for its lead drug candidate, ANB033. The drug, which targets the CD122 receptor to modulate immune response, has demonstrated efficacy in managing the biological symptoms of celiac disease. Following the disclosure, industry analysts have lauded the candidate as a “pipeline-in-a-product,” signaling potential applications far beyond the initial target of celiac, with company shares rising approximately 7% in Monday’s trading session.

The Challenge of Celiac Disease

Celiac disease is a chronic, systemic immune disorder characterized by an adverse reaction to gluten—a protein found in wheat, barley, and rye. When an individual with celiac disease consumes gluten, the immune system mistakenly launches an attack on the small intestine. This process triggers chronic inflammation that, over time, damages the intestinal lining, or villi, leading to malabsorption of essential vitamins and minerals.

For decades, the only effective management strategy has been a strict, lifelong gluten-free diet. However, strict adherence is notoriously difficult to maintain, and many patients continue to suffer from residual symptoms due to accidental gluten exposure. Consequently, the medical community has long viewed celiac disease as a massive, underserved market—the largest autoimmune condition currently without an FDA-approved pharmacological treatment.

Chronology of Clinical Development

The pursuit of a drug-based intervention for celiac has accelerated significantly over the last 18 months, with First Tracks, Argenx, and Teva Pharmaceuticals emerging as the primary contenders.

  • Mid-2025: Interest in the CD122 and IL-15 pathways intensifies as pharmaceutical giants begin identifying these as the “linchpins” of gluten-induced autoimmune damage.
  • Early 2026: Teva Pharmaceuticals reports promising early data from its own proprietary celiac candidate, focusing specifically on IL-15 inhibition.
  • Mid-2026: Argenx, following a strategic acquisition, bolsters its portfolio with a candidate targeting the CD122 receptor, effectively entering a head-to-head developmental race with First Tracks.
  • October 12, 2026: First Tracks releases the results of its Phase 1b trial, confirming that ANB033 meets primary endpoints for safety and biological activity.

Supporting Data: The ANB033 Trial

The trial conducted by First Tracks involved a cohort of 32 participants. The study design was rigorous: participants were randomized into groups receiving either a placebo or three subcutaneous doses of ANB033 administered over a four-week duration. Following the treatment phase, the participants underwent a two-week "gluten challenge," wherein they consumed the equivalent of two slices of bread daily—a standard stress test for celiac therapies.

The data revealed that ANB033 hit statistical significance across three distinct biological markers associated with celiac disease. These markers indicate a reduction in the inflammatory response that typically follows gluten ingestion. Furthermore, the company reported a “clean safety profile,” meaning there were no severe adverse events that would jeopardize the drug’s continued development.

David Risinger, an analyst at Leerink Partners, noted in a client advisory on Monday that the data compares “favorably” to the results recently disclosed by Teva. While the companies have not conducted direct, head-to-head clinical trials, the strength of the biomarkers observed in the First Tracks study suggests a high degree of potency for ANB033.

Official Responses and Strategic Vision

The potential of ANB033 is not limited to celiac disease. Because the drug blocks the activity of CD122—a receptor component shared by the immune-regulating proteins IL-2 and IL-15—it acts as a broad-spectrum immune modulator.

"Celiac disease is the largest autoimmune disease without an approved treatment," said Monique da Silva, Senior Vice President of Corporate Affairs at First Tracks, in an interview with BioPharma Dive. "Should these results be confirmed in larger, pivotal trials, ANB033 could offer patients a new layer of protection that goes beyond the current, restrictive gluten-free diet, significantly improving their quality of life."

The company is already looking toward a diversified future. According to internal projections, First Tracks has outlined an aggressive development roadmap:

  1. Eosinophilic Esophagitis (EoE): A Phase 1b trial is currently underway, with clinical data expected to be unveiled in the third quarter of 2027.
  2. Vitiligo: The company is on track to initiate Phase 2 development for this skin condition in the first half of 2027.
  3. Unnamed Indication: First Tracks has teased a third, as-yet-unnamed autoimmune indication that will enter Phase 2 testing alongside its vitiligo program in the first half of 2027.

Implications for the Future of Autoimmunity

The "pipeline-in-a-product" label assigned to ANB033 is significant. In the pharmaceutical industry, this term is reserved for assets that, if successful in one indication, provide a validated platform to address multiple related diseases. By targeting a fundamental mechanism of immune over-activity, First Tracks is positioning itself to disrupt several segments of the autoimmune market simultaneously.

If the success of the current trial is mirrored in the upcoming EoE and vitiligo studies, the market valuation of First Tracks could shift dramatically. However, the path to commercialization remains complex. Regulatory hurdles, the need for large-scale Phase 3 trials, and the eventual challenge of navigating reimbursement landscapes for chronic autoimmune therapies will be the next major tests for the company.

For now, the investment community remains optimistic. The 7% uptick in share price reflects confidence in the company’s clinical execution and the broader potential of its CD122-blocking technology. Patients, meanwhile, are watching closely; for a condition that has historically been managed only by avoidance, the prospect of a biological "shield" represents a paradigm shift in how celiac disease is understood and treated.

As the industry pivots toward more targeted, mechanism-specific immunotherapies, First Tracks appears to have secured a front-row seat. Whether ANB033 will be the definitive solution for the millions of people living with celiac disease remains to be seen, but the data published this week provides the strongest evidence yet that a medical breakthrough is on the horizon.
